ABSTRACT

Introduction and Objective. From March 2022, China has been in the rapid progressing stage of the Omicron outbreak. However, the mental status of clinical nurses against infection by the Omicron variant of COVID-19 has been not explored. Therefore, a nationwide online investigation with a larger sample size was conducted to explore the mental status of Chinese frontline clinical nurses, and its influencing factors using sound validated and reliable measurements. Materials and method. A cross-sectional study was conducted, and 1,204 clinical nurses fighting the Omicron outbreak were recruited across various provinces of China. Results. The mean age of the nurses was 30.43 (SD=6.59) years. The majority were female. The rates of these nurses with depression, anxiety, stress, and insomnia symptoms were 29.2%, 37.2%, 19.1%, and 48.8%, respectively. The variables of department, insomnia, fear with COVID-19, turnover intention, job burnout, work coping style, and public recognition of nursing, were significant factors influencing the mental status of clinical nurses. Conclusions. The Chinese clinical nurses fighting the Omicron variant of COVID-19 had a similar level of depression and anxiety, a higher level of insomnia, and a lower level of stress, in comparison with nurses globally who experienced the initial period of the pandemic. Targeted investigation and interventions are urgently needed for Chinese frontline clinical nurses with high levels of anxiety, depression and insomnia, who fought against infection by Omicron. Much more professional advocacy is strongly recommended during control of the pandemic and recovery to consolidate the role and influence of nurses. ABSTRACT

Naphthoquinones harboring 1,4-naphthoquinone pharmacophore are considered as privileged structures in medicinal chemistry. In pharmaceutical industry and fundamental research, polyketide naphthoquinones were widely produced by heterologous expression of polyketide synthases in microbial chassis cells, such as Saccharomyces cerevisiae and Escherichia coli. Nevertheless, these cell factories still remain, to a great degree, black boxes that often exceed engineers’ expectations. In this work, the biotransformation of juglone or 1,4-naphthoquinone was conducted to generate novel derivatives and it was revealed that these two naphthoquinones can indeed be modified by the chassis cells. Seventeen derivatives, including 6 novel compounds, were isolated and their structural characterizations indicated the attachment of certain metabolites of chassis cells to naphthoquinones. Some of these biosynthesized derivatives were reported as potent antimicrobial agents with reduced cytotoxic activities. Additionally, molecular docking as simple and quick in silico approach was performed to screen the biosynthesized compounds for their potential antiviral activity. It was found that compound 11 and 17 showed the most promising binding affinities against Nsp9 of SARS-CoV-2, demonstrating their potential antiviral activities. Overall, this work provides a new approach to generate novel molecules in the commonly used chassis cells, which would expand the chemical diversity for the drug development pipeline. It also reveals a novel insight into the potential of the catalytic power of the most widely used chassis cells. ABSTRACT

Objective To study the rule of Chinese medicine prescription According to the prescription novel coronavirus pneumonia;to provide reference for the treatment of epidemic diseases. Methods Through crawling 227 prescriptions of Xinguan TCM collected by Huabing data website intelligent TCM big data platform, we analyzed the web page data by using word cloud analysis, data visualization and the third-party library lxml and request of Python. Results High frequency of drug use of traditional Chinese medicine are: Huoxiang, Atractylodes, Platycodon, honeysuckle, astragalus, Scutellaria, Atractylodes macrocephala, etc. The analysis of clinical symptoms showed that the most common symptoms were fatigue, fever, white fur, cough, chest tightness, diarrhea and so on. Hebei, Sichuan, Heilongjiang, Gansu and other provinces provide more. Conclusion The novel coronavirus pneumonia and almond novel coronavirus pneumonia treatment are better. The results showed that the effective prescriptions and fever, fatigue and other common clinical manifestations, as well as the provinces with higher prescriptions, have important reference significance for the follow-up development of the new crown pneumonia. © 2020 ACM.
